Morris water maze:
The Morris water maze was invented by Morris in 1981 [1]. The method we use is mainly based on [2]. It is divided into five days of positioning navigation test and one day of space exploration test.
Quadrant settings:
At the upper edge of the Morris pool, set 4 mark points in the east, south, west and north at equal distances as the water inlet points of the animal into the pool. 4 equal quadrants.
Platform settings:
According to the experimental requirements, the platform can be arbitrarily set in the middle of a quadrant. Generally, the platform is hidden 1 ~ 2 cm below the water surface. Do not exceed 2cm, preferably 1cm.
Water temperature:
The water temperature is maintained at 22 ° C.
Rat feeding sequence:
Each rat is trained 4 times a day. On the first day, the rats were placed into the pool at the 4 water entry points of east, west, south and north (the head was gently placed toward the wall of the pool). On the second day, follow the directions of west, south, north and east. On the third day, follow the directions of South, North, East, and West. On the fourth day, follow the north, east, west, and south directions. On the fifth day, follow the east, west, south, and north directions. In the sixth day of space exploration experiment, the input point is the opposite side of the quadrant where the platform is located
time setting:
If the animal fails to find the platform in the pool or fails to climb the platform within 90 s of swimming in the water, the tester can place the animal on the platform and stand for 15 s to make the animal feel the feeling on the platform The spatial location of the platform.
After the animal climbs onto the platform, let the animal stand on the platform for 10 s. Remove the rat from the platform and rest for 30 to 60 s before training again.
Precautions:
1. Be gentle when feeding rats to avoid stress. Head up, avoid immersion in water.
2. Injecting rats is done by the same experimenter, and cannot be replaced. And before the formal experiment, touch the rats every day to familiarize them with the smell and avoid stress.
3. When the mouse is placed in the pool, the experiment operator should immediately squat down, and the mouse should not be used as a reference object, because the experimenter is constantly moving inside, which affects the spatial positioning of the mouse.
4. Before the water maze is officially done, put each group of mice into the pool and let it swim for 2 minutes, on the one hand, to let them know that there is no escaped exit in the pool, on the other hand, to eliminate the individual will not Swimming mice so as not to disturb the data behind.
5. Do not turn on the phone, do not speak, try to make gestures to avoid sound interference.
